    The system provides a PCR based high-throughput method for preparing full-sites siRNA library.
  • The method of this kit is easy to practice;
    		•
  • The length of siRNAs constructed by this kit rang from 19-23bp by applying a type III restriction enzyme-Ecop15I and responsible Linkers;
    		•
  • High recombination rate(>80%), and low repeat sequences by random sequencing;
    		•
  • The siRNA expression vector:pU6H1-GFP (size:5.5kb)
    		•
    a. It has dual RNA polymerase III promoters: U6 and H1, can be used to express in mammalian cells.
    b. It contains Green Fluorescent Protein Gene (GFP) as a gene marker, to easy to control the transfection is effective.
    c. Between the start signal "AAAAA" and a termination sequence of RNA polymerase III "TTTTT", there is a pure siRNA sequence, has no any other sequence extra.
  • The method can also be modified to construct microRNA library.
